Restoration reaction of high-pressure oxygen to the injury of vascular endothelium and its function induced by carbon monoxide toxicosis in rat

Aim: To discuss the effects of acute carbon monoxide(CO) toxicosis on vascular endothelium and its cellular functions as well as the therapeutic reaction of high-pressure oxygen in rat. Methods: Carbon monoxide toxic rat model was established according to Jiang's methods. Serous levels of nitric oxide (NO) and endothelin-1 and the numbers of circular endothelial cell were assayed immediately after toxicosis and on the 1st, 3rd, 7th and 14th day respectively with corresponding controls. Results: Serous NO level markedly decreased while endothelin-1 level and circular endothelial cell numbers markedly increased immediately after CO toxicosis; serous NO level markedly increased but endothelin-1 level and circular endothelial cell numbers markedly decreased after high-pressure oxygen therapy in toxic rat. All above index were gradually return to normal levels. Conclusion: CO toxicosis could cause sever functional damages in vascular endothelium and its cells; single or combine index assay of serous NO, endothelin-1 and circular endothelial cell numbers can be used as important managements clinically to judge the extent of damages in vascular endothelium and its functions as well as to evaluate therapeutic effectiveness and prognosis in CO toxic patients.
