AIM: The neuroprotective action of APP17-mer peptide for glutamate toxicity was preliminarily confirmed by observing the effect of APP17-mer peptide and glutamate on growth of SY5Y neuroblastoma cells and expression of NT-3 and NF. METHODS: Measurement of MTT metabolic rate, LDH leakage rate and cell count, Western blot examination of SY5Y expression of NT3 and NF. RESULTS: Glutamate reduced MTT metabolic rate, cell count and expression of NT-3 and NF, but increased LDH leakage rate. The addition of APP17-mer peptide almost or fully normalized the foregoing indicators. CONCLUSION: Glutamate is neurotoxic, and APP17-mer peptide can reduce this damage, and is neuroprotective.
