Aim: To explore the feasibility of repairing minipig mandibular critical-sized defects using mineralized collagen scaffold (MC) combined with in situ periosteal ossification. Materials and Methods: Six minipigs were divided randomly into two groups, with three animals in each group. Segmental bone defect with a length of 6 cm in the right mandible was created and the in situ periosteum was preserved. In Group A, MC was implanted in the defects, while the defects were left empty in Group B. The animals were sacrificed at 12 weeks after surgery. The specimens were evaluated by gross, radiology, histology and bone density examinations. Results: The segmental bone defect was repaired in Group A while it failed in Group B. The bone repair approach in Group A was intramembranous ossification. Conclusions: The critical-sized mandibular defect in minipig can be repaired using MC combined with periosteal ossification, which indicates that MC is an appropriate scaffold for bone tissue engineering.
